چکیده

Microtubules are hollow rod-like polymers (figure 1). They fulfill a varied range of functions including maintenance of cell integrity, forming a spindle that facilitates chromosome separation and as a protein polymer ‘track’ for intracellular transport by molecular motors such as kinesin and dynein. Each microtubule is made of 13 protofilaments consisting of linearly arranged tubulin molecules. Each molecule is a dimer of two closely related monomers α and β tubulin, each having a molecular weight of 50 kDa. Tubulin dimers show micro-heterogeneity on Isoelectric focusing gels (figure 2) in part due to tubulin being encoded by a multi-gene family and in part due to an extensive range of posttranslational modifications. Tubulin’s posttranslational modifications are varied and include acetylation, glutamylation, glycation, phosphorylation and tyrosination. The atomic structure of the tubulin molecule was recently described (figure 3).

Tyrosination of α-tubulin controls the initiation of processive dynein-dynactin motility.

Post-translational modifications (PTMs) of α/β-tubulin are believed to regulate interactions with microtubule-binding proteins. A well-characterized PTM involves in the removal and re-ligation of the C-terminal tyrosine on α-tubulin, but the purpose of this tyrosination-detyrosination cycle remains elusive. Here, we examined the processive motility of mammalian dynein complexed with dynactin an...

Structural basis of tubulin tyrosination by tubulin tyrosine ligase

Tubulin tyrosine ligase (TTL) catalyzes the post-translational retyrosination of detyrosinated α-tubulin. Despite the indispensable role of TTL in cell and organism development, its molecular mechanism of action is poorly understood. By solving crystal structures of TTL in complex with tubulin, we here demonstrate that TTL binds to the α and β subunits of tubulin and recognizes the curved confo...

Tubulin tyrosination is required for the recruitment of CAP-Gly microtubule plus-end- tracking proteins at microtubule ends

Tubulin-tyrosine-ligase (TTL), the enzyme which catalyzes the addition of a C-terminal tyrosine residue to alpha-tubulin in the tubulin tyrosination cycle, is involved in tumor progression and has a vital role in neuronal organization. We show that in mammalian fibroblasts, CLIP-170, and other microtubule tip tracking proteins comprising a CAP-Gly microtubule-binding domain such as CLIP-115 and...

Tubulin tyrosination is a major factor affecting the recruitment of CAP-Gly proteins at microtubule plus ends

Complete separation of tyrosinated, detyrosinated, and nontyrosinatable brain tubulin subpopulations using affinity chromatography.

The maximum achievable tyrosination level of neurotubulin, in vitro, is about 50%. We have developed a method to obtain a complete separation of the tyrosinatable and nontyrosinatable species. We use an immunoaffinity column, with coupled YL 1/2 monoclonal antibody (anti-Tyr-tubulin) and rapid desalting methods. Both subpopulations can be obtained in a polymerizable, apparently native, form. We...
